Roaccutane (isotretinoin) and similar medicines can definitely cause severe birth defects. They must not be taken in pregnancy, and you should wait at least a month after stopping the drug before stopping contraception. The other commonly used drugs-tetracycline antibiotics such as doxycline, should also not be used in pregnancy though data on its adverse effects (staining teeth and affecting bone growth) is pretty sparse.
